    Hi,
    I am using the live cell imaging chamber (model CV-30) for cells visualization and first I seeded the cells on the 30 mm glass coverslip (top) but I couldn’t focus the cells properly, so I tried to seed them on the 40 mm coverslip (bottom) and now I can focus the cells using the 10X objective. The problem is that I cannot focus at all the cells when I try to use the 60X oil-immersion objective. It seems that the sample is too thick for the objective to be able to focus the cells. Am I doing something wrong? Or for this chamber it can only be used a dry objective?

    Hi Paula,

    First you should check which gaskets are being used. Are you assembling the chamber with the 250-um gaskets or the 375-um gaskets? If you are using the thicker gaskets, you will not be able to focus properly. The chamber is intended for use with oil immersion objectives.

    You were right! I mixed by mistake the two gaskets!
